Often times, the users will share the keyboard & mouse. Sometimes the host would only allow voice & gamepad inputs (which is what I recommend, especially if you are going to walk away from the computer).

Edit: It is more preferred as a hot-seat style if mouse & keyboard is required, When considering online play in Tabletop Simulator, you can have some people on that computer and some on another for the same exact game, as long as the total player count is not exceeded (meaning 3 solo computers + 2 shared computers is possible).
